You don't need to be familiar with the game to enjoy the show!
I came into this series only knowing it was adapted from a popular game, but not much else. (And if you're wondering about the 2 in the title, it's not a sequel to any previous anime, its a stand alone series). Fortunately, even though I'm not a gamer, I had no problem following the plot as everything is set up and explained pretty well. This is definitely a "no one is safe" show as each new "trial" that comes up over the seven day period takes heavier and heavier tolls on the characters and the world (you'll notice that when a character dies, they also disappear from the ending animation sequence). The show does start kind of slow but as the story progresses it keeps getting better and the ending wraps it all up pretty well.
On the down side, character development is lacking and the lead character's beliefs and actions aren't well explained, but given the number of episodes and and the fast pacing of the show its somewhat understandable.
